Abstract
The aim of this study was to optimize the yield of polysaccharides and protein, gel viscosity and textural properties of Linum usitatissimum mucilage using a sequential design. The study factors were the extraction method (without agitation, with magnetic stirring and with ultrasound), temperature (40, 55, 70 °C), extraction time (1, 2 and 3 h), pH (5.5, 7.0 and 8.5) and seed:water ratio (1:10, 1:20 and 1:30 g/L). In the first stage, a screening design was used to determine that the temperature (° C) and time (h) of extraction were the factors that have a significant effect on the process. It was evidenced that the extraction method has no significant effect on the yield and percentage of proteins. In the second stage, using a complete design, it was possible to determine that the optimal conditions of extraction of the mucilage were: 85 to 90 °C; 4.5 to 5.0 h; pH 7.0 and seed ratio:water of 1:20, these conditions optimize the yield (9.73%), the percentage of proteins (12.5%) and provide a gel with a viscosity of 1110.90 mPa.s and 159.97 g of firmness, 568.48 g.s of consistency, 29.52 g of cohesiveness and 14.48 g.s of viscosity index.
